The town was incorporated in 1922. The California Trail, Oregon Trail and Mormon Trail passed south of Glendo. The Overland Stage Line also passed south of Glendo.

He was in the cockpit, near the town that William L. Carlisle, one of Americas last train robbers, was captured in December 1919 after a shootout with a detachment in pursuit of him.

On 21 August 2017, Glendo was right in the middle of the path of a total solar Eclipse, the first over the continental United States since 1979. As in many places, the path of totality, thousands of eager tourists and Eclipse hunters rushed in Glendo, to witness the event.
